Force modulation of selectin mediated leukocyte adhesive dynamics

Studies suggest that selectin mediated rolling requires a relatively low reactive bond compliance compared to antigen-monoclonal antibody interactions. Based on in vivo and in vitro evidence, selectin tethers were directly measured at high temporal resolution comparing their kinetics and reactive compliance at similar site densities. L-selectin bonds with neutrophil ligands were observed to be significantly more liable in the presence of shear forces than either of the two vascular selectins. The bond lifetimes were found to have significant differences in reactive compliance, demonstrating that selectin ligands may contribute to different bond lifetimes under stress.
