Statement of Purpose: Dermal fibroblasts (DFs) are resident mesenchymal cells that produce collagen and other extracellular matrix proteins. Cultured autologous dermal fibroblasts are believed to reduce wrinkle formation and promote skin rejuvenation. One clinical application for autologous dermal fibroblasts has been approved by the Food and Drug Administration (FDA). Herein, three-dimensional (3D) spheroids were cultured to increase the therapeutic potential of DFs. In addition, platelets rich plasma (PRP) was used as scaffolds to enhance the viability and retention time of cells after intradermal injection. This study investigated the effects of self-assembled DF spheroids with the combination of PRP on the anti-aging effect by reducing wrinkles on aged nude mice.
