An LC-MS/MS method for determination of glycosaminoglycan content of urine for the diagnosis of mucopolysaccharidoses

A quantitative liquid chromatography/mass spectrometry/mass spectrometry (LC/MS/MS) is developed to measure the characterstic disaccharide derived from glycosaminoglycans (GAG) by partial methanolysis. The results show that methanolysis replace the sulphate groups by hydroxyl groups and methylates. The polymeric GAG collapse to a common disaccharide, the sodiated adduct of which dissociates to a monosaccharide by CID. The measured GAG concentrations were normalized by urinary creatinine content.
