Contributing to consortia human tissue mapping efforts with multiplexed imaging
Consortia projects are harnessing the power of single-cell, spatially resolved imaging technologies for large-scale tissue mapping projects. We have participated in normal and disease tissue mapping consortia by contributing to the development of tools, standards, and multiplexed imaging data. We describe these experiences and contributions and offer advice on the experimental design and practical elements of executing a consortium experiment. We focus on consortia-based considerations for antibody panel creation, array design, metadata collection, and standardization for data sharing. We also describe how these efforts can spur innovations in data processing and analysis and lead to biological insights. Other groups who have recently joined or are considering involvement in tissue-mapping consortia projects, especially those employing multiplexed imaging, can use this chapter as a primer to improve engagement and overall contributions to consortia efforts.
