Enzyme conjugation with polyethylene glycol (PEG) is one method for solving problems in enzyme replacement therapy for metabolic diseases. An intramuscular injection of bovine adenosine deaminase (ADA) conjugated with 5 kDa mono-methoxy PEG was used to treat severe combined immunodeficiency (SCID). Pharmacokinetics were assessed by monitoring plasma ADA activity during treatment, while the immunologic response to PEG-ADA was monitored by ELISA.
