The preparation of cytokine detection protein arrays for in vitro biomaterials evaluation was investigated. The mouse monocytes, RAW 264.7 were plated in 6-well plates at 105 cell/well in 2mL of DMEM medium. Corresponding culture media were collected, and assessed for temporal cytokine content. It was found that the arrays can detect very low cytokine levels directly from cell culture medium without any detectable non-specific cross reactivity.
