The localization and characterization of peptide acylation with ester linkage by electron capture dissociation FTICR/MS were discussed. The electron capture dissociation (ECD) FTICR/MS were applied to ghrelin, a 28-residue acylated growth-hormone-releasing peptide. It was found that the precursor molecular ions isolated by stored-waveform inverse Fourier transform (SWIFT) ejection were irradiated with low energy electrons from the heated EI filament. The structure of the attached acyl group was also deduced from the accurate mass measurement of the fatty acid neutral loss from the molecular ion.
