Isomeric oligosaccharide analysis using hydrophilic interaction LC-ESI-MS/MS

A hydrophilic interaction liquid chromatography electrospray ionization mass spectrometry (LC-ESI-MS/MS) method was established for the separation and analysis of hexose tetrasaccharide linkage isomers in urine, plasma and blood spots. Using the LC-ESI-MS/MS method, the extent of interference from other isomers was also determined. It was found that the tentative identification of isomer 1 as maltotetraose was supported by the increase in both isomer 1 and Glc 4 observed in the plasma of GSD II patients, suggesting it to be derived from glycogen. It was also found that isomer 1 is the major isomer present in blood spot extracts and is probably derived from glycogen stored in the red cells.
